Learnaboutthetechnologiesunderlyingexperimentationusedinsystemsbiology,withparticularfocusonRNAsequencing,massspec-basedproteomics,flow/masscytometryandlive-cellimaging.Akeydriverofthesystemsbiologyfieldisthetechnologyallowingustodelvedeeperandwiderintohowcellsrespondtoexperimentalperturbations.Thisinturnsallowsustobuildmoredetailedquantitativemodelsofcellularfunction,whichcangiveimportantinsightintoapplicationsrangingfrombiotechnologytohumandisease.Thiscoursegivesabroadoverviewofavarietyofcurrentexperimentaltechniquesusedinmodernsystemsbiology,withfocusonobtainingthequantitativedataneededforcomputationalmodelingpurposesindownstreamanalyses.Wedivedeeplyintofourtechnologiesinparticular,mRNAsequencing,massspectrometry-basedproteomics,flow/masscytometry,andlive-cellimaging.Thesetechniquesareoftenusedinsystemsbiologyandrangefromgenome-widecoveragetosinglemoleculecoverage,millionsofcellstosinglecells,andsingletimepointstofrequentlysampledtimecourses.Wepresentnotonlythetheoreticalbackgrounduponwhichthesetechnologieswork,butalsoenterrealwetlabenvironmentstoprovideinstructiononhowthesetechniquesareperformedinpractice,andhowresultantdataareanalyzedforqualityandcontent.
